RELOCATING BONUS AVAILABLE FOR THOSE WILLING TO MOVE OUT OF STATEPrimaryResponsibilities:· Use prior expertise to plan and establish phage library bio panning experiments. · Use different screening strategies to enrich library against target protein · Perform phage-ELISAs to identify binders with different affinities · Sequence positive clones and identify unique sequences · Clone antibody fragments in different vectors using standard molecular cloning techniques (PCR, restriction analysis, ligation, transformation, colony PCR screens) · Generate plasmids expressing scFv, site directed mutants, and fusion proteins using PCR or Gibson assembly. · Engineer expression vectors with insertion or deletion of restriction sites, epitope tags) · Construct phage display libraries, titer and assess diversity · Express and purify antibodies from E.coli or mammalian cells· Perform a variety of assays to characterize antibodies (ELISAs, Western blotting, Flow cytometry). · Prepare plasmids in small/large scale, maintain plasmid and bacterial glycerol stocks. · Prepare bacteriophage stocks for large number of clones and maintain accurate records of all associated data (binding, titer, functional assays). · Perform cell-based (proliferation, cytotoxicity, transcription factor reporter) assays in multi well formats to functionally validate antibodies · Perform experiments with minimal supervision, think critically and demonstrate troubleshooting and problem-solving skills. · Self-motivated, pays attention to details, and adapt to responsibilities outside of the initial job description · Develop precise SOPs complying with GLP environment and maintain excellent lab records that allow systematic tracking and protocol transfer. · Basic proficiency in various software (Excel, PowerPoint, Graph Pad Prism, Photoshop, and Plasmid design/analysis software) Knowledge,Skills & Abilities:Critical to have the ability to maintain accurate records and back track phage clones

Communicate ideas and results clearly and effectively (both verbally and in writing) to the team members.

Must feel comfortable in a fast-paced startup environment with minimal direction and able to adjust workload based upon changing priorities

Ability to adapt to startup environment and make the best use of the existing resources to helping the team to move towards perfection

Familiarity with routine lab operations and safety procedures.

Education& Professional Experience:MS Degree preferably with at least 2+ years of laboratory experience in the field of Immunology, cell biology, virology, or molecular biology.

Experience in phage display library construction, antibody engineering, humanization or affinity maturation of antibodies are preferred .

Experience in molecular cloning and fragment assembly techniques

Experience in setting up cell based and biochemical assays in 96 and 384 well formats.

Experience in working with multiple cell lines (both suspension and adherent)

General lab experience in operating and maintaining basic lab equipment (centrifuge, microscope, PCR machines, nanodrop, DNA and protein electrophoresis systems).

Hands on experience with sterile microbiology and molecular biology techniques not limited to cloning, mutagenesis, restriction digestion, ligation, transformation, colony PCR, culturing bacterial cells, plasmid preparations.

WorkingConditions:· Laboratory environment- required to work with blood products, tumor cell lines, bacteria, retrovirus, lentivirus, and lab chemicals used in the lab with proper Personal Protective Equipment as required · Ability to stand for a period at laboratory benches · Required to retrieve samples from liquid nitrogen and subzero freezers · Must wear PPE due to safety requirements in designated lab areas · May be required to work in controlled or clean room environments · Expected to work safely and effectively when working alone or working with others  RELOCATING BONUS AVAILABLE FOR THOSE WILLING TO MOVE OUT OF STATEAbout Actalent: Actalent connects passion with purpose.
